pluriMate
pluriMate is a new PBMC isolation aid for perfect blood overlay every time. pluriMate dramatically assists the researcher in obtaining quick, consistent results.
pluriMate® was developed for optimal separation of leukocytes and peripheral blood mononuclear cells (PBMC) from whole blood and bone marrow. The key feature of pluriMate® is the porous sponge incorporated at the bottom of the centrifuge tube. This barrier consists of high-grade polyurethane. It prevents you from time-consuming and laborious overlaying of the sample material. Anticoagulated blood or bone marrow can simply be poured directly from the blood sampling tube into the pluriMate® tube. The porous barrier prevents mixture of the sample material with the separation medium. During centrifugation, leucocytes, lymphocytes and PBMCs are separated from unwanted erythrocytes and granulocytes, depending on the density gradient used (Leuko Spin, Lympho Spin, PLT Spin etc.) on the basis of their density and enriched in an interphase above the separation medium. When separation is complete, the barrier prevents recontamination of the enriched cell fraction during harvest. Regardless of whether large or small sample volumes are to be processed.

Bead size and surface modification
pluriBead® is using non-magnetic monodisperse microparticles for the sorting of cell mixtures. The surface of the beads is modified with antibodies that recognize specific structures on the cell surface. The beads are larger than the cells and enlarge these artificially. The enlargement by the cells allows a simple separation through sieves and the use of different separation strategies.
During incubation, the cells in suspension are marked with the help of the particles. The targets bonded to the surface of the catcher particles are then separated from all unbound cells through our sieving system. Finally, the target cells are gently detached from the catcher particles with a detachment buffer.
The size of pluriBead® allows easy and rapid separation of cells from whole blood or buffy coat. Pretreatment of blood and blood components, such as the production of a mononuclear cell fraction, is not required, ie, PBMC ( peripheral blood mononuclear cells)made by a density gradient / density centrifugation or other methods of depletion of erythrocytes (e.g. erythrolysis) or target concentration is not required. Moreover, the particle size prevents uptake by endocytosis. The microbeads are available in various sizes and surface modifications.

A physiologically 3D dynamic culture has been shown to be more sensitive than in a static cell culture. They are also more resistant to drug candidates than a 2D cell culture.
Down regulation of anti-apoptotic features have been observed in physiologically 3D dynamic cultures.
LiveBox systems have been shown to provide a closer prediction of in-vivo drug efficacy than a 2D static model.
The modularity of the IVTech LiveBox culture systems allows greater accuracy in the study of complex tissue responses such as inflammation and cancer.

Tension Systems
Flexcell® Tension Systems are patented, computer-regulated bioreactors that apply cyclic or static strain to cells cultured in vitro. We designed Flexcell® Tension Systems to help investigators analyze biochemical changes in response to tensile load on a variety of cell culture applications, including muscle, lung, heart, vascular vessels, skin, tendon, ligament, cartilage, and bone. Our Tension Systems work with UniFlex® (Uniaxial strain) and/or BioFlex® (Equibiaxial strain) culture plates.
Compression Systems
Flexcell’s® Compression System regulates positive air pressure to compress tissue samples or 3D cell cultures in vitro. The FX-5000C™ Compression System compresses samples between a piston and stationary platen using our BioPress™ culture plates. It can load up to 14 lbs. of applied force. Using the FX-5000C™ Compression System, researchers can observe biochemical changes and cell signaling in vitro that mimics in vivo conditions.
Fluid Shear Systems
The Streamer® is a fluid shear stress device allowing users to regulate fluid shear stress to cells in culture with laminar, pulsatile, or oscillating flow. Flexcell’s® Streamer® System uses a computer-controlled peristaltic pump. Together with the Osci-Flow® Flow Controller, researchers can regulate the frequency of oscillation or pulsatile patterns based on shear stress level applied from 0-35 dynes/cm2. Flexcell’s® Fluid Shear System is designed to allow users to observe biochemical changes, cell migration and signaling under fluid shear load in vitro.
